NHS Magnetic Beads

The functional group on the surface of NHS magnetic beads is NHS, which can couple with proteins or small molecules with primary ammonia to form stable peptide bonds, which can be used for affinity purification of antibodies, antigens, etc. Compared with conventional carboxyl magnetic beads and amino magnetic beads, NHS magnetic beads have the characteristics of simple operation and mild coupling conditions.

1 Product Overview
The functional group on the surface of NHS magnetic beads is NHS, which can couple with proteins or small molecules with primary ammonia to form stable peptide bonds, which can be used for affinity purification of antibodies, antigens, etc. Compared with conventional carboxyl magnetic beads and amino magnetic beads, NHS magnetic beads have the characteristics of simple operation and mild coupling conditions.

4 How to use
Take an appropriate amount of magnetic beads and wash them several times with pH 8.0 buffer solution. Disperse an appropriate amount of biomolecules in a pH 8.0 buffer, redisperse the magnetic beads in this solution, and couple at 25°C for 3 hours or overnight at 4°C.
After coupling, magnetically absorb the beads, discard the supernatant, add buffer containing 1% BSA, redisperse the beads, and couple at 25°C for 1 hour to block unreacted NHS groups.
After blocking, magnetically absorb the beads, remove the supernatant, wash the beads 3 times with 20 mM pH 7.4 PBS buffer, redisperse, and store at 4°C for later use. If necessary, add 0.02% sodium azide to inhibit bacterial growth. .
5 things to note
Magnetic beads have a high density and will easily sink if left standing for a long time. Therefore, please shake well before use to obtain a uniform magnetic bead suspension.
The coupling buffer used during coupling must not contain ammonia substances, otherwise it will affect the coupling efficiency.
When blocking, ethanolamine solution, saturated Tris solution, etc. can be selected for blocking according to the needs of the application. The NHS group is very reactive and sensitive to water. Therefore, after taking the magnetic beads, please seal the remaining magnetic beads with a sealing film and store them at 4°C.
